פרופ' עמיחי מזר נולד בחיפה בשנת 1942, למד בבית הספר התיכון הריאלי בחיפה, ולאחר שירות צבאי בנח"ל החל בשנת 1963 בלימודי ארכיאולוגיה והיסטוריה של עם ישראל באוניברסיטה העברית בירושלים. את מסלול לימודיו האקדמיים עשה באוניברסיטה העברית עד לקבלת התואר השלישי (1976), והשתלם באוניברסיטת לונדון. מאז שנת 1978 הוא שימש כחבר סגל במכון לארכיאולוגיה באוניברסיטה העברית עד לפרישתו מן ההוראה בדרגת פרופ' מן המנין בשנת 2010. במרוצת השנים הוא שימש כמרצה וחוקר אורח באוניברסיטאות פנסילבניה, הארוורד וקליפורניה בסן דייגו והופיע כמרצה אורח באוניברסיטאות וכנסים רבים. הוא היה פעמיים חבר בקבוצות מחקר במכון ללימודים מתקדמים של האוניברסיטה העברית.
עמיחי מזר שימש במשך שנתיים כעוזר ראשי לפרופ' נחמן אביגד בחפירות הרובע היהודי בירושלים (1969-1971). עבודת השדה שלו כללה ניהול סקר אמות המים הקדומות לירושלים (1968), חפירות תל קסילה (1972-1974, 1982-1988), תל בטש (תמנה המקראית), בעמק נחל שורק (1977-1989), תל בית שאן (1989-1996) ותל רחוב בעמק בית שאן (1997-2012). בנוסף לאלו, לעיתים בשותפות עם אחרים, הוא ניהל חפירות קטנות-היקף במספר אתרים מתקופות שונות (שכונת גילה בירושלים, חירת מרג'מה, מירבת אבו טווין, חורבת שילחה, הרטוב, "אתר הפר", חורבת ערש). חפירות אלו שפכו אור על מגוון נושאים בארכיאולוגיה של ארץ ישראל, החל משלהי האלף הרביעי לפנה"ס ועד לתקופה הפרסית.
מחקריו של פרופ' מזר עוסקים בצדדים שונים של התרבות החומרת של ארץ ישראל בתקופות הברונזה והברזל בהקשר הרחב של האזור, וכן קשר בין הממצא הארכיאולוגיה להיסטוריה של תקופת המקרא. הוא פרסם (בשנת 1990) ספר יסוד בארכיאולוגיה של א"י בתקופת המקרא. כתב וערך תשעה ספרים בהם פורסמו תוצאות החפירות המרכזיות שלו (תל קסילה, תל בטש, תל בית שאן), פרסם מאמרים רבים במגוון נושאים בארכיאולוגיה של תקופת המקרא, ובהם מספר מאמרי סינתיזה מקיפים.
שימש כראש המכון לארכיאולוגיה, עורך כתב העת "קדמוניות" במשך חמש שנים וכאחד מעורכי כתב העת Israel Exploration Jornal וכן היה עורך וחבר מערכת של מערכת של מספר קבצי מאמרים.
פרופ' מזר זכה בפרס ישראל למחקר בארכיאולוגיה (2010), בפרס לנדאו של מפעל הפיז ופרס שימל המוענק על ידי מוזיאון ישראל. בשנת 2006 יצא כרך מאמרים לכבודו. הוא חבר האקדמיה הישראלית למדעים.
נשוי לאורה ולהם שלושה ילדים וחמישה נכדים.
מחקר פעיל
מחקר ממצא ופרסום מדעי של חפירות תל רחוב. עריכת שישה כרכים של פרסומי החפירה בשיתוף עם ד"ר נאוה פניץ-כהן וקבוצת חוקרים נוספים. הפרסום צפוי לצאת לאור בשנת 2019 בסדרה "קדם" QEDEM של המכון לארכיאולוגיה.
